Also known as: E433, Polyoxyethylene (20) sorbitan monooleate, Tween 80

Synthetic nonionic surfactant/emulsifier widely used in processed foods, ice cream, and sauces. Peer-reviewed work links dietary emulsifiers (including polysorbate 80 and the related emulsifier CMC) to gut-microbiota alteration and intestinal inflammation in animal models. EFSA re-evaluated E433 in 2015 (group ADI 25 mg/kg bw/day).

Concern

Two commonly used emulsifiers, carboxymethylcellulose and polysorbate-80, induced low-grade inflammation and obesity/metabolic syndrome in wild-type mice and promoted robust colitis in mice predisposed to this disorder, via alteration of the gut microbiota.

In its 2015 re-evaluation, the EFSA ANS Panel allocated a group ADI of 25 mg/kg bw/day to polysorbates E432, E433 (polysorbate 80), E434, E435 and E436, derived from a rat carcinogenicity NOAEL of 2,500 mg/kg bw/day with an uncertainty factor of 100.

EFSA's 2015 re-evaluation allocated a group ADI of 25 mg/kg bw/day to the polysorbates including E433 (polysorbate-80), from a rat NOAEL of 2,500 mg/kg bw/day with a 100x uncertainty factor.
